The river usually flows from south to south southeast to south southwest, combining sixteen tributaries , including the Little And Timbarra , And flows into Lake King, one of the main lakes in the vast system of Gippsland lakes. Within the lake, the Tambo River forms a confluence with the Mitchell River, west of the village of Metung , which flows into the Bass Strait , southwest of Lakes Entrance Lakes Entrance, in the county of East Gippsland county East Gipsland . The river descends 1,060 meters (3,480 feet) along its 186-kilometer (116 miles) watercourse [3] .

The river leaves the mountains and the picturesque nature reserve of Mount Tambo, forming its headwaters near the village of Bindi, north of the Swift Creek, and flows from Bindi through the Tambo Valley to Buten. A large alpine road takes the river route to Tonga and follows the river for most of the distance through the Tambo Valley south of this point. the valley from Bindi south of Ensay is relatively open and flat and inhabited as agricultural land, however it closes again to become a steep wooded mountain beyond Ensay.
