Son of Berardo da Varano , nephew of Gentile da Varano, whom he succeeded in 1355.
In 1344 he participated in the capture of Smyrna , in 1355 he defeated Galeotto Malatesta under Paderno di Ancona and Castelfidardo . In the same year, he concluded a peace treaty with Galeotto and later married his daughter to him.
Since 1360, he commanded papal forces in the capture of Rimini, Fano, Pesaro, Fossombrone, Ascoli Piceno and Forlì. Later, by condotta, he served the Kingdom of Naples , Governor of Abruzzo .
In 1362, on the side of Florence, participated in the war with Pisa, captured Pechioli.
In 1370, he fought for the Florentines with Bernabo Visconti , in 1377 - on the contrary, for Pope Gregory XI against Florence.
Since 1355, the lord Camerino , also in his possession included the cities of Macerata and Numana , the towns (castra) of Tolentino and San Ginesio .
He died in Tolentino in 1384.

After the death of Rodolfo II, the war for inheritance began. In 1386, an agreement was concluded according to which Cameroino was received by his brother Gentile III and the son of the last Rodolfo III, and the sons of another brother, Venanzio, who died in 1377 - Gentire and Berardo, formerly expelled from the city, could return with the return of confiscated property.
