Functions
The enzyme catalyzes the addition of fucose through an alpha-1,6-glycosidic bond of the 1st residue of N-acetylglucosamine , which is closest to the N-glycan peptide chain.
Structure and intracellular localization
FUT8 consists of 575 amino acids , a molecular weight of 66.5 kDa. Like other fucosyltransferases , it is localized on the membranes of the trans-section of the Golgi apparatus .
Expression
Expressed in almost all tans, the highest level of expression is in the nerve plexus of the corpus callosum of the brain.
Pathology
Mutations of the fucosyltransferase 8 gene can lead to a disease of a hereditary glycosylation disorder with defective fucosylation (so-called CDGF). In connection with the multiple functions of protein glycosylation, the disease is characterized by a wide range of clinical manifestations: defects in the development of the nervous system, psychomotor retardation, dysmorphisms, hypotension, coagulation disorders and immunodeficiency. CDGF is an autosomal recessive disorder.
