XbaI is a restriction endonuclease isolated from the bacterium Xanthomonas badrii [1] .

Properties
XbaI refers to restriction endonucleases (Type II, subtype P). That is, XbaI recognizes the palindromic DNA sequence and carries out restriction within the palindrome at a fixed point. In the presence of dimethyl sulfoxide (DMSO) and at high glycerol concentrations in the medium, XbaI loses its ability to recognize a palindrome and performs non-specific restriction ( stellar activity ) . After restriction, sticky ends with protrusions from four bases are formed.

XbaI restriction activity is inhibited by methylation of adenine ( DAM methylase ). However, restriction enzyme activity persists upon methylation of cytosine ( DCM or CpG methylase) .
After DNA restriction is complete, XbaI can be inactivated by denaturation by heating to 65 ° C for 20 minutes .
Application
XbaI is widely used in molecular biological research. In particular, XbaI is used for cloning specific nucleotide sequences [2] , in restriction analysis .
For example, XbaI was used in the analysis of polymorphism of estrogen receptor α [3] and lipoprotein a (ApoB) [4] .
