Trim Away Method

The Trim-Away method is a method of rapid forced highly selective (selective) degradation of endogenous proteins in mammalian cells , which does not require preliminary modification of the genome or matrix RNA . Since the selectivity of the Trim-Away method is based on the use of antibodies , it can be applied to a wide range of target proteins using ready-made monoclonal antibodies . Trim-Away allows you to study the function of the protein in a variety of cell types [1] [2] . Designed by Melina Schuh in collaboration with Dean Clift [3] .

Principle of Operation

The method is based on the use of a ubiquitin ligase , TRIM21 , which binds with high affinity to the domain of a crystallizing fragment of immunoglobulin Fc antibodies [4] . During infection, TRIM21 transports the ubiquitin proteasome system to antibody-associated pathogens, leading to their destruction.
