Throughout the sixteenth century, the Tsutsui clan sought to subjugate Yamato province to its power. The most famous is Tsutsui Junkey (1549-1584). In 1577, Junkei, being on the side of Oda Nobunaga , defeated Matsunagu Hisahide at the battle of Sigisan. As a reward, Oda Nobunaga handed over Yamato to Zenkei. After the death of Oda Nobunaga in 1582, Tsutsui Junkey arrived at the site of the battle of Yamazaki and waited for someone to win, after which he attacked the forces of the defeated Aketi Mitsuhide . The new ruler Toyotomi Hideyoshi retained Yamato province for Tsutsui Junkei, but reduced his income.

In 1584, after the death of Junkey, the head of the Tsutsui clan became his nephew and adopted son Tsutsui Sadatsugu (1562-1615). In that 1584, by order of Toyotomi Hideyoshi Tsutsui, Sadatsugu was forced to transfer the province of Yamato Toyotomi Hidenage , and he was transferred to Iga province , where he built the new Iga Ueno castle. In 1600, Tsutsui Sadatsugu opposed Uesugi Kagekatsu on the side of Tokugawa Ieyasu , but was later deprived of possession for abuse of power.

During the Edo period, representatives of the Tsutsui clan did not play a big role.
