Puromycin

Puromycin is a purine-derived antibiotic produced by Streptomyces alboniger bacteria. It is an inhibitor of protein synthesis . In chemical structure, puromycin is similar to aminoacyl tRNA . It readily interacts with the A center of peptidyl tRNA to form peptidyl puromycin. The latter does not carry an anticodon triplet and thus inhibits the elongation of the growing peptide chain, causing premature termination of protein synthesis. Puromycin has an equally effective inhibitory effect on protein synthesis in both prokaryotes and eukaryotes . Due to this, puromycin is used as an antibiotic against gram-positive and some gram-negative bacteria, protozoa (trypanosomes, amoeba), a number of helminths (pinworms, tapeworms). It is also widely used in cell biology as a selective agent for various kinds of cell cultures expressing the Puro marker (puromycin-N-acetyl transferase).
