Cyclin D1 is a protein from the cyclin family that specifically regulates the G1 / S phase transition in the cell cycle. In humans, it is encoded by the CCND1 gene located at chromosome locus 11q13. [1] [2]

Function
Cyclin D1 belongs to the family of evolutionarily very conserved proteins - cyclins . Members of this family are characterized by a sharp dependence of the content of these proteins in the cell on the phase of the cell cycle. Cyclins function as regulators of the so-called cyclin-dependent kinases (CDKs). Different cyclins exhibit different patterns of expression and degradation in different phases of the cell cycle. In the complex, all this contributes to strict temporal coordination of the work of various intracellular signaling cascades during each mitotic event and the correct course of the cell cycle. So, in particular, cyclin D1 forms a complex with cyclin-dependent kinases CDK4 and CDK6, and serves as a regulatory subunit of these kinases (activates them). In turn, the cyclin D1 / CDK4 complex or cyclin D1 / CDK6 activates (phosphorylates) the so-called Rb retinoblastoma protein (tumor suppressor), which contributes to the progression of the cell cycle and the transition from G1 to S phase. And the expression of the cyclin D1 gene, in turn, is positively regulated by the Rb protein. Mutations, amplification, and overexpression of the cyclin D1 gene, accelerating cell cycle progression and cell division, are often observed in various tumors and may contribute to oncogenesis (tumor development).
Immunohistochemical staining for cyclin D1 is used to diagnose mantle cell lymphoma .
It was also found that cyclin D1 is overexpressed in breast cancer cells . Its use as a potential biomarker has been proposed. [3]
