CD154 , also a CD40, CD40L ligand, or TNFSF5 ( English Tumor necrosis factor ligand superfamily member 5 ) is a cytokine of the tumor necrosis factor family, mainly localized on activated T-lymphocytes . Functions
TNFSF5 (often referred to as CD40L ) is part of a large family of tumor necrosis factors ( TNF ). CD40L plays the role of a costimulatory molecule and induces the activation of antigen-presenting cells during stimulation of the T-cell receptor by the molecules of the main histocompatibility complex located on antigen-presenting cells. Interacts with CD40 and integrins α5β1 ( VLA-5 ) and αIIbβ3 ( glycoprotein IIb / IIIa ).
CD154 is expressed on T lymphocytes . The protein regulates the function of B-lymphocytes by interacting with CD40 on the surface of B-cells.
Structure
Protein exists in two forms: membrane-bound and soluble. The membrane-bound form consists of 261 amino acids, and the molecular weight is 29.3 kDa. Cleavage of the N-terminal fragment (112 amino acids) releases protein into the medium. Contains 1 N-glycosolation site and one intramolecular disulfide bond.
It is a homotrimer.
Pathology
Gene disruptions lead to the inability to switch immunoglobulin classes and are associated with high IgM syndrome. [one]
