Functions
The NOD2 gene is a member of the NOD1 / Apaf-1 subfamily, or Nod-like receptors containing the CARD domain. Expressed mainly by peripheral blood leukocytes . It plays a role in the immune response to bacterial products carrying muramyl dipeptide and activates the pro-inflammatory transcription factor NF-κB . [2] Also involved in transplant rejection reactions.
Structure
Mature protein consists of 1,040 amino acids , and the molecular weight is 115.3 kDa . The molecule includes two CARD domains, a NACHT domain, 9 LRR ( leucine enriched) repeats, and an ATP binding site .
Binds to RIPK2 / RICK via interaction between protein CARD domains.
Tissue specificity
Expressed in monocytes .
In pathology
Mutation of the NOD2 gene leads to Crohn's disease and Blau's syndrome.
