N-cadherin

Functions

N-cadherin together with E- and P-cadherins refers to the 1st type of cadherin, calcium- dependent proteins of cell adhesion. They are involved in homophilic interactions, forming cell-cell contacts. N-cadherin is involved in the mechanism of neuronal recognition. In hippocampal neurons, it can regulate the density of dendritic spines . [2]

Structure

N-cadherin consists of 747 amino acids (after maturation), the molecular weight of the protein part is 99.8 kDa . The main N-terminal site (565 amino acids) is extracellular, then there is a single transmembrane fragment and an intracellular fragment (161 amino acids). The extracellular fragment includes 5 repeats of the cadherin domain and from 4 to 7 N-glycosylation sites . The cytosol fragment contains a phosphorylation site (on the tyrosine residue).

Interacts with CDCP1. Detected in conjunction with FGFR4, NCAM1 , PLCG1, FRS2, SRC, SHC1, GAP43 and CTTN. Interacts with PCDH8, forming a complex that can contain TAOK2 and internalize through a mechanism mediated by TAOK2 / p38 MAPK.
