To participate in the tournament, football associations of 7 countries that agreed to take part in the tournament were presented with one team each - the champion of the last 1959 . The exception was Brazil, because the championship has not yet been held in this country. Brazil was represented by the Bahia Cup winner from Salvador . However, the Confederation of Football of Brazil in 2010 equated the Brazil Cup (1959-1968) to the national championship.
- Argentina - San Lorenzo de Almagro ( Buenos Aires ) (champion of Argentina 1959 )
- Bolivia - Jorge Wilstermann ( Cochabamba ) ( 1959 Bolivian Champion)
- Brazil - Bahia ( Salvador ) ( Brazil Cup Winner 1959 )
- Colombia - Millionarios ( Santa Fe de Bogota ) ( 1959 Colombian Champion)
- Paraguay - Olympia ( Asuncion ) (champion of Paraguay 1959 )
- Uruguay - Peñarol ( Montevideo ) (champion of Uruguay 1959 )
