Periciazine

Periciazin ( lat. Periciazinum ) - an antipsychotic agent , belongs to the group of piperidine derivatives of phenothiazine [1] . This group of drugs has less potent antipsychotic activity than aliphatic and piperazine derivatives of phenothiazine [2] .

General Information

Like other antipsychotics, pericyazine potentiates the effects of drugs, sleeping pills and analgesics . It has adrenolytic and relatively strong anticholinergic activity; has a strong antiemetic effect [2] .

The antipsychotic effect of periciazine is combined with a sedative without a stimulating component [2] .

The drug is indicated in the treatment of patients with both psychotic and psychopathic conditions. It is especially effective for behavioral disorders (especially in children), contact disorders. It reduces aggressiveness and, thanks to a milder sedation than chlorpromazine , does not cause severe lethargy, lethargy, and is well tolerated [2] .

Due to a more selective, than other antipsychotics, normalizing effect on behavior, periciazine is called in the literature β€œbehavior corrector” [2] .

Prescribe periciazine orally after meals. The dose for adults is first 5-10 mg per day; then it is gradually increased (every 1–2 days by 10 mg) until the optimal effect is obtained [2] (usually up to 30–40 mg [2] , and in some cases up to 70–90 mg per day).

The daily dose is given in 2-3 doses: in the morning 1 / 4-1 / 3 of the daily dose, the rest - in the evening (due to the possible development of drowsiness) [2] .

Children and seniors are prescribed starting from 5 mg per day, then the dose is gradually increased to 10-20-30 mg per day. After obtaining a persistent effect, the dose of the drug is gradually reduced, selecting an individual maintenance dose [2] .

Side Effects

Depression, spasmodic torticollis , oculomotor crises, trismus , other extrapyramidal disorders , orthostatic hypotension , constipation, dry mouth [3] , urinary retention, accommodation paralysis, hyperthermia [2] , amenorrhea , impotence , galactorrhea , weight gain [3] , photosensitization , allergic reactions [4] , extremely rarely - cholestatic jaundice , agranulocytosis [3] .

Contraindications

A history of toxic agranulocytosis, a closed-angle form of glaucoma , porphyria , prostate disease [3] .

Precautions

With caution is prescribed for diseases of the liver, kidneys and cardiovascular system , as well as elderly patients. When prescribing periciazine to patients suffering from epilepsy (the drug can lower the threshold of convulsive readiness) and parkinsonism , especially careful medical supervision is necessary. If hyperthermia develops, periciazine should be discontinued [3] .

With caution, prescribe the drug to drivers and people working with mechanisms, because of the possibility of drowsiness, especially at the beginning of therapy. During the use of periciazine is prohibited to drink alcohol [3] .

Drug Interactions

It enhances the effect of antihypertensive and hypnotics , tranquilizers , painkillers [3] , drugs for general anesthesia , and alcohol - containing drugs. With the combination of periciazine with paroxetine , the effect of both drugs is enhanced. When combined with sedatives or hypnotics or with procarbazine , the sedation is enhanced [5] .
